目的

探讨青蒿琥酯(Artesunate,ART)对乳腺癌MCF-7细胞株的增殖、分化功能及细胞形态和结构的影响。

方法

ART作用乳腺癌MCF-7细胞后,采用MTT(四唑盐)比色法检测细胞增殖功能,观察细胞形态和结构的改变,流式细胞仪分析细胞周期相分布及细胞凋亡率。统计学分析采用重复测量设计资料的方差分析。

结果

ART对乳腺癌细胞MCF-7有抑制作用,随浓度增加和时间延长抑制作用增强(P<0.05),呈浓度依赖及时间依赖,并可导致细胞形态和结构的改变,抑制MCF-7细胞增殖,阻滞MCF-7细胞于S期和G2/M期。6μmol/L和8 μmol/L ART诱导MCF-7细胞的凋亡率分别为3.15%和8.43%。

结论

ART可导致MCF-7细胞形态的改变,抑制MCF-7细胞增殖和生长,阻滞MCF-7细胞于S期和G2/M期,并有诱导细胞凋亡的作用。

Objective

To study the effect of artesunate(ART)on proliferation and morphology of breast cancer MCF-7 cells.

Methods

After MCF-7 cells were treated with ART,MTT-assay was performed to examine the proliferation of MCF-7 cells and observe their morphological changes,and FCM(flow cytometry)assay was used to analyze the distributions of cell cycles and cell apoptosis.Statistical analysis was done using repeated measures analysis of variance.

Results

ART had suppressive effect on MCF-cells and with increase of the concentration and time,the suppressive effect was enhanced(P<0.05),showing dose-and timedependent.ART could inhibit the proliferation of MCF-7 cells,change the morphological structure of MCF-7 cells,and arrest MCF-7 cells in the S and G2/M phases of cell cycle.ART of 6 μmol/L and 8 μmol/L could cause 3.15%and 8.43%apoptosis of MCF-7 cells,respectively.

Conclusions

ART has the capability of changing morphological structure,inhibiting proliferation of MCF-7 cells and arresting MCF-7 cells in S and G2/M phases in cell cycle.It also induces apoptosis of MCF-7 cells.
