目的

了解雌激素受体β1（ERβ1）对雌激素敏感性指状蛋白（Efp）基因的调节作用，为进一步探讨ERβ 对Efp基因的调控机制奠定基础。

方法

应用脂质体法将ERβ1真核表达质粒转染至乳腺癌MCF-7细胞中， 再用Western blot检测转染细胞中Efp蛋白表达的变化。 MTT比色试验观察ERβ1真核表达质粒转染后MCF-7细胞增殖活性的变化。

结果

外源性ERβ1真核表达质粒组MCF-7细胞较未转染组MCF-7细胞Efp蛋白表达明显减弱。 ERβ1基因转染后的MCF-7 细胞的增殖活性降低。

结论

ERβ1基因的表达可以抑制人乳腺癌MCF-7 细胞中Efp基因的表达， 并抑制MCF-7 细胞的增殖能力，可能在乳腺肿瘤发生、发展机制中具有重要的作用。

Objective

To explore the effects of exogenous ERβ1 gene on the expression of estrogenresponsive finger protein （Efp） in MCF-7 cell line.

Methods

pcDNA3.1-ERβ1 plasmid was transfected into MCF-7 cell line by lipofectin reagent. The expression of Efp was determined by Western Blot. The influence of ERβ1 gene on proliferation of breast cancer MCF-7 cell was detected by MTT test.

Results

The expression level of Efp was obviously lower in transfected MCF-7 cell than that in the control. Exogenous ERβ1 could inhibit proliferation of MCF-7 cell.

Conclusions

ERβ1 gene may inhibit the expression of Efp gene and proliferation of MCF-7 cell. It plays an important part in the carcinogenesis of breast cancer.
